Chondrocyte Migration is Modulated by Osmotic Environment
ثبت نشده
چکیده
INTRODUCTION In situ osmolarity of cartilage tissue is estimated to be 350-450 mOsm [1-2]. In joint injury and arthritis, breakdown of tissue macromolecules and increasing water content is estimated to result in a hypotonic environment (~300 mOsm), which can also be measured from the synovial fluid [3-4]. Many studies have investigated the influence of extracellular osmolarity on chondrocytes, which are usually cultured in medium of ~300 mOsm [5-7]. We previously reported the effects of medium osmolarity on chondrocyte cytoskeleton organization and hydraulic permeability [8-9]. In the current study, we hypothesize that extracellular osmotic conditions modulate chondrocyte migration.

Effects of Osmotic Loading on Chondrocyte Nuclear Size
Chao, P G; Bulinski, J C; Ateshian, G A; +Hung, C T Columbia University, New York, NY [email protected] INTRODUCTION Cartilage compression changes cell shape, volume and hydrostatic and osmotic pressure of chondrocytes in situ [1,2]. For example, changes in the osmotic environment of cells have been shown to modulate chondrocyte biosynthesis, however, the mechanosensing mechanisms are not well...
متن کاملEffect of Long-Term Osmotic Loading Culture on Matrix Synthesis from Intervertebral Disc Cells
The intervertebral disc is a highly hydrated tissue that acts to absorb and distribute large complex loads placed on the spine. Diurnal loading and disc degeneration causes significant changes in water volume and proteoglycan content, which alters the internal osmotic environment. Short-term osmotic loading alters disc cell gene expression; however, the long-term effect of osmotic loading on di...
متن کاملEffect of osmotic stress on the expression of TRPV4 and BKCa channels and possible interaction with ERK1/2 and p38 in cultured equine chondrocytes.
The metabolic activity of articular chondrocytes is influenced by osmotic alterations that occur in articular cartilage secondary to mechanical load. The mechanisms that sense and transduce mechanical signals from cell swelling and initiate volume regulation are poorly understood. The purpose of this study was to investigate how the expression of two putative osmolyte channels [transient recept...
متن کاملMeasuring principles of frictional coefficients in cartilaginous tissues and its substitutes.
The frictional properties of cartilaginous tissues, such as the hydraulic permeability, the electro-osmotic permeability, the diffusion coefficients of various ions and solutes, and the electrical conductance, are vital data to characterise the extracellular environment in which chondrocytes reside. This paper analyses one-dimensional measurement principles of these coefficients. Particular att...
متن کاملComparing Behavior of Chondrocyte Cells on Different Polyhydroxybutyrate Scaffold Structure for Cartilage Tissue Engineering
Introduction: As the ability to repair cartilage tissue in body is limited, finding a suitable method for cartilage regeneration has gained the attention of many scholars. For this purpose, scaffold structure and morphology, along with cell culture on it, can be a novel method to treat cartilage injuries, osteoarthritis. Methods: In this study, polyhydroxybutyrate (PHB) is selected as the scaf...
